ABSTRACT

Objetivou-se comparar o efeito in silico do florfenicol nas doses de 20 e 30 mg/Kg em ovinos pelas vias intravenosa (IV) e intramuscular (IM), usando a modelagem PK/PD. Realizou-se uma simulação de Monte Carlo com base nos dados de concentração plasmática de um estudo publicado anteriormente. Calculou-se a área sob a curva (ASC) e as taxas de eficácia do florfenicol para os efeitos bacteriostático, bactericida e de erradicação bacteriológica. A dose de 20 mg/Kg IV demonstrou efeitos de erradicação de 100, 93 e 0% para CIM de 0,5, 1 e acima, respectivamente. O efeito bacteriostático foi de 99 e 90% para CIM de 4 e 2 µg/ml, enquanto o bactericida foi de 14% para CIM de 2 µg/ml. A dose de 30 mg/Kg IV apresentou 100% de erradicação para CIM de 1 µg/mL e 100% de efeito bactericida para CIM de 2 µg/mL. Há 100% de efeito bacteriostático em CIM de 4 µg/ml. As doses de 20 e 30 mg/Kg IM mostraram 100% de erradicação para CIM até 1 µg/mL e 0% para CIM maiores. O efeito bacteriostático foi mantido em 100% para uma CIM de 4 µg/mL em ambas as doses. Este estudo mostra o efeito de erradicação bacteriológica do florfenicol nas doses de 20 e 30 mg/Kg, IV e IM. Recomenda-se que seja feito um estudo de eficácia in vivo com a dose de 30mg/Kg IM em ovinos infectados por F. necrophorum com MIC superior a 2 µg/mL.


We aimed to compare the in silico effect of florfenicol at doses of 20 and 30 mg/Kg in sheep by intravenous (IV) and intramuscular (IM) routes, using PK/PD modeling. We performed a Monte Carlo simulation based on plasma concentration data from a previously published study. We calculated the area under the curve (AUC) and the efficacy rates of florfenicol to bacteriostatic, bactericidal, and bacteriological eradication effects. The dose of 20 mg/Kg IV demonstrated 100, 93, and 0% eradication effects for MICs of 0.5, 1, and above, respectively. The bacteriostatic effect was 99 and 90% for MIC of 4 and 2 µg/ml, while the bactericide was 14% for MIC of 2 µg/ml. The 30 mg/Kg IV dose showed 100% eradication for MIC of 1 µg/mL and 100% bactericidal effect for MIC of 2 µg/mL. There is a 100% of bacteriostatic effect at MIC of 4 µg/ml. Doses of 20 and 30 mg/Kg IM showed 100% eradication for MIC up to 1 µg/mL and 0% for MIC above. The bacteriostatic effect was maintained at 100% for a MIC of 4 µg/mL at both doses. This study shows the bacteriological eradication effect of florfenicol at doses of 20 and 30 mg/Kg, IV, and IM. Therefore, we recommend an in vivo efficacy study with a dose of 30mg/Kg IM in sheep infected with F. necrophorum with MIC greater than two µg/mL.

ABSTRACT

OBJECTIVE: To evaluate the effects of heel-opened ankle foot orthosis (HOAFO) on hemiparetic gait after stroke, especially on external foot rotation, and to compare the effects of HOAFO with conventional plastic-AFO (pAFO) and barefoot during gait. METHODS: This cross-over observational study involved 15 hemiparetic patients with external rotation of the affected foot. All subjects were able to walk independently, regardless of their usual use of a single cane, and had a less than fair-grade in ankle dorsiflexion power. Each patient was asked to walk in three conditions with randomized sequences: 1) barefoot, 2) with a pAFO, and 3) with an HOAFO. Their gait patterns were analyzed using a motion analysis system. RESULTS: Fifteen patients consisted of nine males and six females. On gait analysis, hip and foot external rotation were significantly greater in pAFO (-3.35degrees and -23.68degrees) than in barefoot and HOAFO conditions (p<0.05). Wearing an HOAFO resulted in significant decreases in hip (0.78degrees, p=0.04) and foot (-17.99degrees, p<0.01) external rotation compared with pAFO; although there was no significant difference between HOAFO and barefoot walking. Walking speed and percentage of single limb support were significantly greater for HOAFO than in barefoot walking. CONCLUSION: HOAFO was superior to pAFO in reducing hip and foot external rotation during the stance phase in patients with post-stroke hemiparesis. HOAFO may, therefore, be useful in patients with excessive external rotation of the foot during conventional pAFO.

ABSTRACT

Neste artigo realizou-se a avaliação de ovinos mestiços Santa Inês com pododermatite infecciosa, verificando as alterações no leucograma e proteínas de fase aguda. No primeiro experimento, 70 fêmeas foram separadas em três grupos de acordo com o seu escore podal: Grupo controle (G1) com escore 0; Dermatite interdigital (G2) com escore 1 ou 2 e Pododermatite necrosante (G3) com escore 3, 4 ou 5. Durante dois meses observou-se a evolução clínica e efeitos no leucograma em cinco momentos: M1 (dia 0), M2 (dia 15), M3 (dia 30), M4 (dia 45) e M5 (dia 60). A intensidade do resultado do leucograma foi de baixa magnitude, observando-se alterações significativas (p<0,05) como uma leve leucocitose (G3, M4), atribuída a neutrofilia e um discreto aumento no número total de monócitos (G3, M2 e M3) apesar do extenso dano e necrose tecidual existentes na última fase da doença. No segundo experimento, utilizaram-se 105 animais de sete propriedades com objetivo de isolar o agente etiológico e avaliar os efeitos da doença sobre proteína plasmática total e proteínas de fase aguda. Os animais também foram separados em três grupos: controle, dermatite interdigital e pododermatite necrosante. Em todas as propriedades foi realizado o isolamento de Dichelobacter nodosus. Não houve correlação significativa (p<0,05) das diferentes fases da doença sobre as proteínas estudadas, porém a haptoglobina dos grupos com animais doentes apresentou médias superiores ao grupo controle. De acordo com a metodologia utilizada e resultados obtidos, conclui-se que as proteínas de fase aguda estudadas não foram eficientes na caracterização das fases da pododermatite infecciosa ovina e que a resposta leucocitária foi branda, tornando difícil sua utilização para este fim.


In this paper, Santa Inês crossbred sheep with footrot were evaluated, checking the changes on leukogram and the acute phase proteins. In the first experiment, 70 females were divided into three groups according to their podal scores: a control group (G1) with score 0, scald group (G2) with score 1 or 2 and a footrot group (G3) with score 3, 4 or 5. During two months, the clinical course and its effect on leukogram were observed at five moments: M1 (day 0), M2 (day 15), M3 (day 30), M4 (day 45) and M5 (day 60). The intensity of the white blood cell count was low magnitude, observing significant changes (p <0.05) as a mild leukocytosis (G3, M4), attributed to neutrophilia and a slight increase in the total number of monocytes (G3, M2 and M3) despite the extensive damage and tissue necrosis existed during the final stage of the disease. In the second experiment, 105 animals from seven farms were used with the objective of isolating the causative agent and to evaluate the effect of disease on total plasma protein and acute phase proteins. The animals were also divided into three groups: control, scald and footrot. In all farms, the isolation of Dichelobacter nodosus was regarded. For the different stages of the disease on the proteins studied there was no significant correlation (p<0.05) but the haptoglobin in scald and footrot groups showed higher average than the control group. Based on these results, we conclude that the acute phase proteins studied were not efficient in the characterization of the phases of the disease and the leukocyte response was mild, making it difficult to use for this purpose.

ABSTRACT

A ocorrência de miíases cutâneas foi verificada em 10 diferentes criatórios de ovinos no norte de Minas Gerais, Brasil, durante o período de um ano, onde foram analisados os fatores relacionados a essas parasitoses. Os rebanhos possuíam predominantemente animais mestiços Santa Inês, criados em sistemas semiextensivo. Foram registrados 50 casos e a maior incidência foi observada nos meses de março (22%) e abril (18%), períodos de maiores temperaturas e umidade relativa do ar. As patas foram as regiões mais frequentemente acometidas (34% dos casos). As lesões por pododermatites (38% dos casos), onfaloflebites (10%), linfadenite (6%) e dermatobiose (6%) foram os fatores mais frequentemente relacionados às miíases. As taxas de ocorrência não diferiram quanto ao sexo e idade dos animais. Foi constatado que a maioria dos proprietários não realizava o tratamento das miíases de forma correta, sendo que após a implantação de uma terapia preconizada nesta pesquisa, 92% das lesões apresentaram cura entre sete e trinta dias. Os resultados apontam a importância de estratégias para prevenção baseadas nos fatores relacionados neste estudo. A inspeção constante dos ovinos, identificando-se os ferimentos e tratando-se as lesões precocemente, devem constituir práticas rotineiras, principalmente no período chuvoso e quando estão presentes nas patas dos animais.


Cutaneous myiasis was investigated in 10 sheep herds during one year in northern Minas Gerais, Brazil, and factors related to its occurrence were analyzed. The sheep were predominantly hybrid Saint Inês, bred in semi-extensive systems. A total of 50 cases were registered and the highest incidence occurred during March (22%) and April (18%), months with high temperatures and high relative humidity of the air. There was no influence of sex and age for the occurrence of myiasis. The lesions were mostly found on the legs (34% of the cases). Pododermatitis (38% of the cases), omphalophlebitis (10%), lymphadenitis (6%) and dermatobiosis (6%) were the predisposing factors more frequently associated with myiasis. The myiasis was not correctly treated by the owners, but after implantation of a correct therapy, 92% of the lesions presented cure within 7 to 30 days. The results show the importance of preventive strategies based on the predisposing factors identified in this study. The constant inspection, identifying wounds and precociously treating the lesions, should be routine especially at the end of rainy season and when predisposing factors are present.

ABSTRACT

This paper reports the occurrence and epidemiology of outbreaks of foot rot and other foot diseases in goats and sheep in the semiarid region of Paraíba, northeastern Brazil. Four farms were inspected for the presence of foot lesion in sheep and goats and for environmental conditions, general hygiene, pastures, and disease control measures. The prevalence of foot lesions was 19.41 percent (170/876) in sheep and 17.99 percent (52/289) in goats, ranging between 5.77 percent and 33.85 percent in different farms. Foot rot was the most common disease, affecting 12.1 percent of the animals examined (141/1165), but with significantly higher (p<0.05) prevalence in sheep (13.69 percent) than in goats (7.27 percent). The frequency of malignant foot rot was also significantly lower (p<0.05) in goats (9.53 percent) than in the sheep (40.83 percent). On one farm, Dorper sheep showed significantly higher (p<0.05) prevalence of foot rot (17.5 percent) than Santa Inês sheep (6.79 percent), and the number of digits affected was also higher in the former. Dichelobacter nodosus and Fusobacterium necrophorum were isolated from cases of foot rot. White line disease was found in 3.95 percent of the animals, sole ulcers in 1.29 percent, foot abscess in 1.03 percent and hoof overgrowth in 0.5 percent. The high rainfall at the time of occurrence, grazing in wetlands, clay soils with poor drainage, presence of numerous stony grounds, closure of the flocks in pens at night, and introduction of affected animals were considered predisposing factors for the occurrence of foot diseases.


Este trabalho relata a ocorrência e aspectos epidemiológicos de surtos de pododermatite infecciosa e outras afecções podais em caprinos e ovinos no semiárido paraibano. As propriedades foram inspecionadas quanto à presença de problemas podais e aspectos relacionados às condições ambientais, higiene, locais de pastejo e medidas de controle utilizadas. Nos ovinos a prevalência de lesões podais foi de 19,41 por cento (170/876) e nos caprinos de 17,99 por cento (52/289), variando entre 5,77 por cento e 33,85 por cento nas diferentes propriedades. A pododermatite infecciosa foi a doença mais frequente acometendo 12,1 por cento dos animais examinados (141/1165), sendo a prevalência nos ovinos (13,69 por cento) significativamente maior (p<0,05) do que a nos caprinos (7,27 por cento). A frequência de lesões malignas desta doença em caprinos (9,53 por cento) foi, também, significativamente menor (p<0,05) do que nos ovinos (40,83 por cento). Em uma das propriedades a prevalência de pododermatite infecciosa entre os ovinos Dorper (17,5 por cento) foi significativamente maior do que a nos ovinos da raça Santa Inês (6,79 por cento), tendo os animais pela pododermatite infecciosa. Dichelobacter nodosus e Fusobacterium necrophorum foram isolados de casos de pododermatite infecciosa. A doença da linha branca foi constatada em 3,95 por cento dos animais, a úlcera de sola em 1,29 por cento, o abscesso do pé em 1,03 por cento, e crescimento excessivo do casco em 0,5 por cento. Os altos índices pluviométricos na época de ocorrência, o pastejo em áreas úmidas, os solos argilosos com pouca drenagem, os terrenos pedregosos, o encerramento em currais durante a noite e a introdução de animais doentes foram considerados fatores predisponentes para a ocorrência dos surtos.

ABSTRACT

An outbreak of footrot has occurred in Debrezeit swine farm that is located 44 km south east of the capital. Among 24 pigs, 75% showed lameness, hot, painful and swollen feet, hemorrhagic bruising of the coronary band, heel erosion, sole ulcers and separation of skin from hoof on hind limbs. Bacteriological examination of the specimen from the affected limbs has shown the involvement of Staphylococcus, Streptococcus and Dichelobacter nodosus. The overt clinical lameness and inability to stand or mount can cause reduced reproductive performance. This preliminary finding thus warrants further epidemiological investigation.
